You can't use the same printer profile for different label templates. I had to create a new identical printer profile while I was creating a new template even tho all of the settings were identical (the existing printer profile was not available via the drop-down menu). Seems like an oversight - usually you're using the same printer for all your labels so you should be able to reuse the profile.
This is by design. Profiles are unique to a template/printer combination. (In DB language, profile-template-printer form a unique key.) Each type of label will most likely need adjusting for each printer it is printed on. For example: An Avery 5160 will need different tweaks to print correctly on printer A than the Brodart 55459003 (or even the Demco WS14942260) will on the same printer. Likewise, the Avery 5160 will probably need different adjustments for use on printer B than on printer A. We could certainly remove this requirement, but I wonder how often this is an issue? I'd like to have a wider discussion before changing it. Maybe we can ask for some feedback on the user list.
